SOB Broth is a nutrient-rich liquid medium used for cultivation of E. coli and for transformation workflows, including competent-cell preparation and post-transformation recovery. Its peptide, vitamin, and defined salt composition provides osmotic support suitable for high-density growth and handling of chemically competent cells. For selection, appropriate antibiotics may be added as required.

  • 2.0% Tryptone
  • 0.5% Yeast Extract
  • 10mM Sodium Chloride
  • 2.5mM Potassium Chloride
  • 10mM Magnesium Chloride
  • 10mM Magnesium Sulfate

  • Post-Transformation Recovery of E. coli: Outgrowth medium for chemically or electrocompetent cells to maximize transformation efficiency.

  • High-Efficiency Cloning Workflows: Supports rapid growth of recombinant E. coli during plasmid cloning and library construction.

  • Plasmid Maintenance and Amplification: Cultivation of plasmid-bearing E. coli for high-yield DNA preparation.

  • Phage Propagation: Facilitates M13 and related filamentous phage infection and amplification in E. coli hosts.

  • Preparation of Competent Cells: Growth medium for E. coli prior to competency induction protocols.

  • SOB Agar Applications: When solidified, used for antibiotic selection and colony growth of E. coli.
