Salmonella-Shigella (SS) Agar Plates are selective and differential agar plates used for the isolation and preliminary differentiation of Salmonella and Shigella from clinical, food, and environmental samples. The medium inhibits many commensal enteric organisms while differentiating lactose fermenters from nonfermenters and indicating hydrogen sulfide production.

Typical colony characteristics: lactose-fermenting organisms produce pink to red colonies; non–lactose fermenters form colorless to translucent colonies. Salmonella often appears as colorless colonies with black centers due to H2S production, while Shigella typically forms colorless colonies without H2S.

Recommended use: streak plates and incubate aerobically at 35–37 °C for 18–24 h. Heavily contaminated specimens may benefit from enrichment prior to plating. Suspect colonies should be confirmed with appropriate biochemical and serological tests.

Format: ready-to-use poured plates, intended for in vitro microbiological use by trained personnel.

  • Salmonella-Shigella Agar
  • Clinical Stool Diagnostics: Isolation and differentiation of Salmonella and Shigella from fecal specimens using Salmonella-Shigella Agar.

  • Food Safety Testing: Screening raw and ready-to-eat foods (meat, poultry, dairy, produce) for Salmonella contamination.

  • Water and Environmental Monitoring: Detection of Salmonella and Shigella in potable/recreational water and environmental swabs.

  • Selective Recovery After Enrichment: Plating from selenite or tetrathionate enrichment broths to recover Salmonella.

  • Differentiation by Lactose Fermentation: Separation of non-lactose fermenters (Salmonella, Shigella) from lactose-fermenting coliforms like Escherichia coli.

  • H2S Production Detection: Identification of H2S-positive colonies (black centers) typical of many Salmonella spp.

  • Public Health Surveillance: Routine screening and outbreak investigations for enteric pathogens Salmonella and Shigella.

  • Quality Control in Pharma/Biotech: Environmental and process monitoring programs targeting Salmonella.

  • Colony Isolation for Confirmation: Purified colony growth for downstream serotyping, biochemical ID, MALDI-TOF, or PCR of Salmonella/Shigella.
