RIPA Buffer Solution is a laboratory lysis buffer used for total protein extraction from cultured cells and tissues. It disrupts cellular and organelle membranes and solubilizes cytoplasmic, membrane-associated, and nuclear proteins for analytical workflows.

The combination of ionic and nonionic detergents, physiological salt, and buffering components supports protein solubilization and pH control during lysis, while a chelating agent limits divalent cation–dependent enzymatic activity. Lysates prepared with this buffer are commonly used for SDS-PAGE, Western blotting, and immunoprecipitation.

  • 150mM Sodium Chloride
  • 1.0% Triton X-100
  • 1.0% Sodium Deoxycholate
  • 0.1% Sodium Dodecyl Sulfate
  • 50mM Tris-HCl
  • 2mM Ethylenediaminetetraacetic acid (EDTA)
  • Cell and Tissue Lysis: Rapid extraction of total protein from cultured cells and tissues using a stringent ionic/non-ionic detergent mix.

  • Membrane and Nuclear Protein Solubilization: Efficient recovery of membrane-bound and nuclear proteins for downstream analyses.

  • Western Blot Sample Preparation: Generation of denatured protein lysates compatible with SDS-PAGE and immunoblotting.

  • Stringent Immunoprecipitation (IP): Lysis and high-stringency washing to reduce nonspecific binding in IP workflows.

  • Chromatin Immunoprecipitation (ChIP) Lysis/Washes: Harsh lysis and washes for ChIP protocols to minimize background.

  • Phosphoprotein Studies: Preparation of lysates for phosphorylation analysis when supplemented with protease/phosphatase inhibitors.

  • Bacterial Cell Lysis: Lysis of organisms such as Escherichia coli with additional enzymatic or mechanical disruption as needed.
