20X Wash Buffer A is a concentrated phosphate-buffered saline with potassium, formulated at high ionic strength and supplemented with a nonionic surfactant. It is intended for routine wash steps in ELISA, immunoassays, Western blotting, and general microplate and membrane workflows. When diluted 1:20 with deionized water, it yields a 1X working solution with near-neutral pH.

Directions: Mix thoroughly before use. Dilute 1 part concentrate with 19 parts deionized water. Store tightly closed. If salt crystallization occurs during storage, warm and mix until fully dissolved.

  • 2.7M Sodium Chloride
  • 54mM Potassium Chloride
  • 86mM Disodium Phosphate
  • 28mM Monopotassium Phosphate
  • 2% Tween-20

  • Western Blotting: Washes PVDF/nitrocellulose membranes between antibody incubations to reduce nonspecific binding.

  • ELISA: Serves as a plate wash buffer between coating, blocking, and antibody steps to lower background.

  • Immunohistochemistry/Immunocytochemistry: Washes fixed tissues or cells during antibody staining workflows.

  • Immunofluorescence Microscopy: Removes unbound fluorophore-conjugated antibodies from coverslips for clearer signal.

  • Flow Cytometry: Washes stained cell suspensions to eliminate unbound antibodies and reduce carryover.

  • Bead-Based Assays/Immunoprecipitation: Rinses magnetic or agarose beads to minimize nonspecific interactions.
