Mannitol Salt Agar Plates are a selective and differential solid medium used for the isolation and differentiation of staphylococci. A high sodium chloride concentration favors the growth of salt-tolerant organisms such as Staphylococcus spp., while other flora are typically inhibited. Mannitol serves as a fermentable carbohydrate, and phenol red is included as a pH indicator.

Mannitol fermentation by organisms such as Staphylococcus aureus produces acid that shifts the indicator from red to yellow, distinguishing mannitol-fermenting colonies from non-fermenters. Proteose peptone and beef extract supply nitrogen, vitamins, and growth factors, and agar provides a firm surface for colony isolation.

  • Selective Isolation of Staphylococci: High 7.5% Sodium Chloride selects for salt-tolerant staphylococci from mixed clinical, food, and environmental samples.

  • Differentiation by Mannitol Fermentation: 1.0% D-Mannitol with 25µg/mL Phenol Red detects acid production (yellow zones) by fermenters.

  • Presumptive Identification of Staphylococcus aureus: Mannitol-positive colonies with yellow medium suggest coagulase-positive S. aureus.

  • Distinguishing Staphylococcus aureus from Staphylococcus epidermidis: S. aureus ferments mannitol (yellow), while S. epidermidis typically does not (red/pink).

  • Nasal Carriage Screening: Swab screening for Staphylococcus aureus (including presumptive MRSA) using 7.5% NaCl selectivity and mannitol/indicator response.

  • Food and Beverage Quality Control: Detection and enumeration of staphylococci in high-salt or processed products.

  • Cosmetics and Personal Care Testing: Routine monitoring for halo‑tolerant staphylococci in finished products and raw materials.

  • Environmental Monitoring: Surface and air sampling (swabs/settle plates) in healthcare and pharma facilities to detect staphylococcal contamination.

  • Confirmation from Enrichment Broths: Plating enriched samples onto MSA to confirm mannitol-fermenting staphylococci.

  • General Cultivation of Salt-Tolerant Gram-Positive Cocci: Proteose Peptone (1.0%) and Beef Extract (0.1%) support growth; 1.5% Agar provides firm medium for colony isolation.
