Mueller Hinton II Broth, Cation Adjusted, is a nutrient medium used for cultivation of non-fastidious bacteria and for standardized antimicrobial susceptibility testing. Defined levels of Ca2+ and Mg2+, along with controlled pH, make it suitable for disk diffusion, broth microdilution, and MIC determinations.

Typical test organisms include Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Enterococcus faecalis. This broth is the liquid counterpart to Mueller Hinton agar in routine susceptibility workflows.

  • 1.75% Acid Hydrolysate of Casein
  • 0.30% Beef Extract
  • 0.15% Soluble Starch
  • Antimicrobial Susceptibility Testing (AST): Standard medium for broth microdilution/macrodilution and disk diffusion inoculum prep; cation adjustment (Ca2+/Mg2+) supports reproducible MICs, especially for aminoglycosides, tetracyclines, and polymyxins.

  • MIC/MBC Determination: Used to measure inhibitory and bactericidal endpoints for clinical isolates.

  • Cultivation of Nonfastidious Bacteria: Supports robust growth of Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Enterococcus faecalis.

  • Quality Control Testing: Employed with ATCC reference strains for media performance and antibiotic potency QC.

  • Antibiotic Screening and Lot Release: Used to evaluate activity of new compounds and verify batch-to-batch consistency.

  • Growth Kinetics and Inoculum Preparation: Generates standardized cell densities for downstream assays and time-kill studies.

  • Supplemented AST for Fastidious Organisms: Serves as a base for supplemented media (e.g., HTM) for testing Haemophilus influenzae and certain streptococci.
