LA Plates with 1.6% Skim Milk and 2% Agar are based on a Luria-style nutrient agar containing tryptone, yeast extract, and sodium chloride, solidified with agar. The medium is supplemented with skim milk to supply casein and lactose as protein and carbohydrate components.

This formulation is used in workflows that examine proteolytic activity and casein utilization by bacterial isolates. It is compatible with routine cultivation and screening of laboratory strains, including E. coli, in qualitative assessments of protein degradation on solid media.

  • 1.00% Tryptone
  • 0.50% Yeast Extract
  • 0.05% Sodium Chloride
  • 2.00% Agar
  • 1.60% Skim Milk
  • Protease Activity Screening: Detect extracellular proteases via casein hydrolysis (clear halos) using the skim milk in the LA base.

  • Differentiation of Proteolytic Bacteria: Assess protease production by Bacillus spp., Pseudomonas aeruginosa, Staphylococcus aureus, and Serratia marcescens.

  • Dairy and Food Microbiology QC: Enumerate and identify proteolytic contaminants in milk and dairy process samples.

  • Mutant and Strain Screening: Select and compare protease-overproducing or protease-deficient bacterial mutants.

  • Enzyme Regulation Studies: Monitor secreted protease expression under different growth conditions or quorum signals (e.g., in Pseudomonas aeruginosa).

  • General Cultivation: Grow and isolate diverse bacteria on a non-selective LA base supplemented with skim milk and 2% agar.
