M9 Minimal Salts Agar Plates with 1% Glucose, Proline-250, Uracil-250 is a defined agar medium for cultivation and selection of bacterial strains on a minimal carbon source. The addition of 1% glucose provides a fermentable carbon and energy source, while proline (250 µg/mL) and uracil (250 µg/mL) fulfill specific auxotrophic requirements in strains deficient in proline or pyrimidine biosynthesis.

This medium is suitable for growth and colony isolation of laboratory strains such as E. coli K-12 derivatives under minimal conditions, with thiamine, magnesium, and calcium included to supply essential cofactors for central metabolic processes.

  • Auxotroph Maintenance: Routine propagation of proline- and uracil-auxotrophic strains (e.g., pro−, pyr− mutants) supported by 250 µg/mL proline and 250 µg/mL uracil.

  • pyrF/5-FOA Workflows: Uracil-supplemented minimal plates for pyrF-based counterselection using 5-FOA in Escherichia coli and related enterobacteria.
