LB Agar Plates with 1% Glucose and Ampicillin-100 is a nutritionally balanced agar medium formulated for the growth and selection of ampicillin-resistant E. coli strains. The addition of 1% glucose supports carbohydrate metabolism and biomass formation, and 100 µg/mL ampicillin sodium salt enables selection of plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast extract
  • 1.0% Sodium chloride
  • 1.0% Cerelose Anhydrous Dextrose (Glucose)
  • 1.5% Agar
  • 100µg/mL Ampicillin Sodium Salt
  • Antibiotic Selection of Transformants: Recovery and selection of ampicillin-resistant Escherichia coli after chemical or electroporation transformation.

  • Plasmid Maintenance/Propagation: Stable maintenance of AmpR plasmids in Escherichia coli on LB agar with 100 µg/mL ampicillin.

  • Reduced Basal Expression During Cloning: 1% glucose minimizes leaky expression from lac/T7-lac promoters, aiding cloning of toxic inserts in Escherichia coli.

  • Colony Isolation and CFU Enumeration: Isolation of single colonies and counting of ampicillin-resistant colony-forming units.

  • Colony PCR/Screening: Picking colonies for PCR, restriction analysis, or sequencing to verify constructs.

  • Library Plating: High-density plating of combinatorial, mutagenesis, or genomic libraries on 150 mm plates.

  • Replica Plating: Transferring selected colonies to secondary selective conditions or induction plates.

  • Strain Archiving and Picking: Picking verified clones for liquid culture inoculation and glycerol stock preparation.

  • Conjugation/Mating Selections: Selecting AmpR plasmid recipients or transconjugants in Escherichia coli.

  • Contamination Control: Suppressing Amp-sensitive contaminants while maintaining desired AmpR Escherichia coli clones.
