LB Agar with carbenicillin-100 and kanamycin-50 is a nutrient agar medium used for cultivation and selection of E. coli strains carrying corresponding antibiotic resistance markers.

The inclusion of 100 µg/mL carbenicillin disodium salt and 50 µg/mL kanamycin sulfate permits selection of transformants maintaining plasmids encoding beta-lactam and aminoglycoside resistance, respectively.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Carbenicillin Disodium Salt
  • 50µg/mL Kanamycin Sulfate
  • Dual-Antibiotic Selection: Selects recombinant E. coli carrying both bla (carbenicillin/ampicillin) and kan (kanamycin) resistance markers.

  • Post-Transformation Screening: Plates heat-shock or electroporation reactions to isolate double-resistant colonies.

  • Dual-Plasmid Maintenance: Propagates co-transformed E. coli for co-expression and multi-vector workflows under simultaneous selection.

  • Colony Picking and Archiving: Enables picking, restreaking, and preparing glycerol stocks of verified double-resistant clones.

  • Colony PCR and Sequencing Prep: Provides colonies for downstream verification of inserts from dual-selected transformants.

  • CFU Enumeration Under Selection: Quantifies transformation efficiency and library diversity with carbenicillin and kanamycin pressure.

  • CRISPR and Genome Editing Workflows: Selects strains carrying editing and helper plasmids with AmpR and KanR markers.

  • Conjugation/Transposition Selection: Recovers E. coli recipients with kanamycin-marked insertions while maintaining carbenicillin-selectable plasmids.
