40% Glucose Solution is an aqueous solution of D‑glucose (dextrose) at 400 g/L. It is supplied ready to use as a carbon source for preparation and supplementation of microbiological and cell culture media. The solution typically exhibits a pH within the USP range for dextrose solutions.

Use to adjust carbohydrate levels in complex or defined media for organisms such as E. coli and Saccharomyces cerevisiae, or for feed additions in microbial cultivation. Add aseptically to cooled, sterilized media to reach the desired final concentration; for example, adding 5 mL of 40% stock to 100 mL of medium yields 2% (w/v) glucose. Typical working ranges are 0.2–2% (w/v), as dictated by the application and organism.

Handling and storage: store at 2–8 °C; do not freeze. Protect from light. Mix gently before use to resuspend any crystallization. Do not autoclave; maintain sterility via aseptic technique. Discard if turbidity, precipitate that does not redissolve, or color change is observed.

  • Catabolite Repression Control: Used to repress catabolite-sensitive promoters (e.g., lac/ara systems) to reduce basal expression in bacterial expression strains.

  • Antibiotic Production and Selection Media: Provides a defined carbon source in specialized media for cultivating and selecting bacteria, including Staphylococcus aureus.

  • Enzymatic and Metabolic Assays: Source and standard for glucose oxidase/hexokinase assays and metabolic studies.

  • Osmolality and Carbon Flux Adjustment: Rapidly tunes culture osmolality and energy supply without large volume changes.
