8 M Guanidine HCl, 50 mM Tris-HCl is a chaotropic, Tris-buffered solution for protein and nucleic acid workflows. The high guanidine concentration disrupts hydrogen bonding and denatures proteins, while Tris-HCl maintains a stable pH during sample processing.

Applications include cell lysis, dissociation of protein–protein and protein–nucleic acid complexes, solubilization of inclusion bodies, and purification of DNA and RNA. The solution may be diluted or removed prior to downstream enzymatic steps, desalting, or chromatography as required by the workflow.

  • Protein Denaturation and Unfolding: Powerful chaotrope for complete protein denaturation prior to analysis or refolding studies.

  • Inclusion Body Solubilization: Solubilizes aggregated proteins from Escherichia coli inclusion bodies to enable downstream purification.

  • Nucleic Acid Purification: Promotes DNA/RNA binding to silica during cleanup and extraction while inactivating nucleases.

  • Cell Lysis and Inactivation: Rapid chemical lysis of bacterial (e.g., E. coli), yeast, and mammalian cells with concurrent protein and nuclease denaturation.

  • RNase/DNase Inactivation: Preserves nucleic acids by inactivating degradative enzymes during isolation workflows.

  • Protein Refolding Workflows: Enables controlled dilution/refolding from 8.0M denaturant for recovery of native structure and activity.

  • Chromatography Elution/Cleaning: Chaotropic elution of tightly bound proteins and regeneration/cleaning of affinity and ion-exchange resins.

  • Endotoxin Disruption: Disrupts LPS interactions to aid endotoxin reduction during recombinant protein preparation.

  • Immunoprecipitation Elution: Elutes antigen–antibody complexes under denaturing conditions for downstream analysis.

  • Sample Preparation for Mass Spectrometry: Denatures proteins to improve digestion efficiency and proteomic analysis.
