Fluid Thioglycollate Medium is a reducing broth used for the cultivation and detection of aerobic, microaerophilic, and obligate anaerobic bacteria. It creates an oxygen gradient that allows evaluation of organism oxygen requirements by the position of growth in the tube, and includes a redox indicator that signals exposure to oxygen at the surface. It is commonly used for sterility testing and for recovering a broad range of bacteria from clinical, environmental, and pharmaceutical samples.

Suitable organisms include obligate anaerobes such as Clostridium spp., facultative anaerobes such as Escherichia coli, and aerobes such as Staphylococcus aureus and Pseudomonas aeruginosa. Typical use involves incubation at 30–35 °C, with minimal agitation to preserve the gradient.

  • Sterility Testing (USP <71>/EP 2.6.1/JP): Detects aerobic, anaerobic, and microaerophilic contaminants in parenterals, ophthalmics, and medical devices using 2.98% Fluid Thioglycollate Medium; supports organisms like Clostridium sporogenes, Staphylococcus aureus, and Pseudomonas aeruginosa.

  • Growth Promotion and Suitability Tests: Verifies medium performance and method suitability for sterility assays with compendial strains such as Clostridium sporogenes, Bacteroides fragilis, and Staphylococcus aureus.

  • Anaerobe Cultivation and Enrichment: Supports recovery of oxygen-sensitive and fastidious organisms from clinical, environmental, and bioburden samples, including Clostridium spp. and Bacteroides fragilis.

  • Bioburden Testing: Used as a recovery medium for microbial load assessment of raw materials, pharmaceutical waters, and in-process samples, facilitating growth of stressed organisms like Escherichia coli and Pseudomonas aeruginosa.

  • Membrane Filtration Sterility Methods: Incubates filters and rinse fluids to detect contaminants after filtration of products, aiding detection of Clostridium spp. and facultative anaerobes.

  • Medical Device and Implant Testing: Incubates device extracts and rinses to reveal microbial contamination by anaerobes and facultative organisms such as Staphylococcus aureus and Pseudomonas aeruginosa.
