10X Borate Saline Buffer, pH 8.3, is a concentrated buffer used to prepare a 1X borate–saline working solution for biochemical and molecular biology workflows. It provides a mildly alkaline environment and ionic strength suitable for sample dilution, reagent preparation, and wash steps.

Upon 1:10 dilution with deionized water, the buffer maintains pH control via borate and conductivity via saline, supporting procedures that require stable pH near 8.3 and consistent ionic conditions.

  • 100mM Boric Acid
  • 1.5M Sodium Chloride
  • Antibody Conjugation and Labeling: Ideal amine-free buffer at pH 8.3 for NHS-ester dye/biotin coupling to antibodies; dilute 10X (100mM Boric Acid, 1.5M Sodium Chloride) to 1X for optimal ionic strength.

  • Immunoassay Diluent and Wash Buffer: Used as a 1X diluent/wash in ELISA and other immunoassays to maintain stable pH 8.3 without Tris interference.

  • Protein Immobilization and Bead Coupling: Supports covalent attachment of proteins to activated resins or surfaces at mildly alkaline pH.

  • Glycoprotein and Carbohydrate-Related Workflows: Borate interacts with cis-diols, aiding in workflows involving glycoproteins and carbohydrates.

  • Enzyme Reactions Sensitive to Primary Amines: Provides a consistent pH 8.3 environment where Tris would inhibit amine-reactive chemistries.

  • Microarray and Slide Printing: Used for spotting/printing antibodies and proteins onto slides while maintaining structure and activity at 1X ionic strength.

  • General Sample Preparation and Storage: Serves as a 1X buffer for protein/nucleic acid handling where stable pH 8.3 and physiological ionic strength are required.
