Ammonium bicarbonate buffer is a volatile, aqueous buffering solution commonly used in proteomics and analytical workflows. It provides buffering capacity in the neutral to mildly basic range and is compatible with enzymatic protein digestion, peptide cleanup, and LC–MS sample preparation.

The volatile ammonium counterion facilitates downstream drying without nonvolatile salt residue. Typical applications include preparing and diluting protein samples, adjusting pH during digestion protocols, and rinsing or equilibrating chromatographic components where a volatile buffer is required.

  • 50mM Ammonium Bicarbonate
  • Proteomic Digestions: Buffer for in-solution and in-gel trypsin digestion of proteins using 50 mM Ammonium Bicarbonate, providing near-neutral pH and volatility for MS compatibility.

  • LC-MS/MS Sample Prep: Volatile buffer for peptide preparation and cleanup, minimizing non-volatile salts prior to mass spectrometry.

  • Solid-Phase Extraction (SPE): Used during peptide desalting and elution steps where a volatile buffer is required for downstream MS.

  • Buffer Exchange/Cleanup: Exchange into 50 mM Ammonium Bicarbonate before lyophilization or MS to facilitate complete removal by drying.

  • Reduction/Alkylation Workflows: Matrix for DTT reduction and IAA alkylation prior to enzymatic digestion in proteomics.

  • Peptide Mapping: Standard buffer for bottom-up proteomic workflows to generate peptide maps compatible with LC-MS analysis.

  • Enzymatic Assays: Provides controlled, near-neutral pH for protein-modifying enzymes where a volatile buffer is advantageous.

  • HPLC/LC Mobile Phase Additive: Employed at low concentrations as a volatile buffering component for peptide separations in LC-MS.

  • Sample Reconstitution: Reconstituting dried peptide/protein digests in 50 mM Ammonium Bicarbonate before further processing.

  • Lyophilization-Friendly Formulations: Preparing samples in a fully volatile buffer to enable residue-free drying and improved MS sensitivity.
