DNase I (RNase-Free), ≥2000 U/mL, is a bovine pancreatic endonuclease that nonspecifically cleaves single‑ and double‑stranded DNA to oligonucleotides with 5′‑phosphate and 3′‑hydroxyl termini. The RNase‑free specification supports handling of RNA-containing samples where ribonuclease contamination is controlled.

Applications include removal of contaminating DNA from RNA preparations, degradation of DNA in cell lysates, pretreatment of samples for RT‑qPCR and RNA‑seq, and elimination of DNA templates in protein or enzyme preparations.

Activity is defined as the amount of enzyme that catalyzes the release of acid‑soluble nucleotides from DNA under standard conditions; the product concentration is ≥2000 U/mL for accurate volumetric dosing.

  • ≥2000U/mL DNase I
  • RNA Purification: Removes contaminating genomic DNA during RNA isolation (on-column or in-solution) using ≥2000U/mL DNase I to preserve RNA integrity.

  • RT-qPCR Preparation: Treats RNA prior to cDNA synthesis to prevent gDNA amplification artifacts; apply ≥2000U/mL DNase I for reliable Ct values.

  • qPCR/RT-qPCR Controls: Generates no-RT and DNase-treated controls to confirm DNA-free RNA inputs using ≥2000U/mL DNase I.

  • Viscosity Reduction in Lysates: Degrades DNA in crude cell lysates (e.g., Escherichia coli) to reduce viscosity and improve clarification and filtration.

  • Protein Purification: Prevents DNA-mediated aggregation and improves chromatography performance by digesting nucleic acids in lysates with RNase-free DNase I.

  • Plasmid and Host DNA Removal: Eliminates residual DNA from viral vector, phage, and protein preparations to meet downstream purity requirements.

  • DNase I Footprinting: Maps protein–DNA interaction sites via controlled, partial digestion for high-resolution footprinting assays.

  • DNase-Seq and Hypersensitivity Assays: Identifies open chromatin regions through selective cleavage at accessible DNA sites.

  • Library Prep Cleanup: Removes unwanted DNA templates prior to RNA-focused library construction to reduce background reads.

  • Cell-Free Systems: Clears contaminating DNA from in vitro transcription/translation reactions without introducing RNase activity.

  • Exosome/EV Preparations: Digests extracellular DNA in vesicle isolates to distinguish internal nucleic acids from surface-bound contaminants.

  • Microbial Sample Processing: Decreases DNA viscosity in lysates from Escherichia coli and other bacteria to enhance centrifugation and filtration efficiency.
