Tris-buffered saline with Tween-20 (TBST), pH 8.0, is a buffered salt solution containing a nonionic surfactant for routine washing and dilution steps in protein blotting and immunoassays. It is used in Western blotting, ELISA, and immunohistochemistry workflows, including antibody incubation, membrane washing, and sample preparation.

At pH 8.0, the Tris buffer provides stable buffering capacity in the near-physiological range, and sodium chloride maintains ionic strength suitable for protein handling. The inclusion of Tween-20 provides gentle detergent action to aid in removal of unbound reagents during wash steps.

  • Western Blotting: Standard wash buffer for membranes to minimize nonspecific binding during antibody incubations.

  • ELISA: Plate wash buffer and antibody diluent to reduce background and improve signal-to-noise.
