Tryptic Soy Agar Settle Plates are pre-poured plates containing enzymatic digests of casein and soybean meal with sodium chloride and agar. The medium is a general-purpose nutrient base suitable for the cultivation of a broad range of nonfastidious microorganisms under aerobic conditions.

These plates are intended for passive air exposure studies using the settle plate method and for surface or personnel monitoring by contact or swab transfer. Representative organisms that can be cultivated on this medium include Escherichia coli, Staphylococcus aureus, Bacillus subtilis, and Pseudomonas aeruginosa.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • Environmental Monitoring (Passive Air/Settle): Placed open in cleanrooms and controlled environments to capture airborne microorganisms for routine monitoring.

  • Aseptic Process Simulation (Media Fills): Used during simulation runs to assess airborne contamination risk during critical operations.

  • Trend Analysis of Microbial Fall-Out: Enables longitudinal trending of ambient bioburden for quality control and release decisions.

  • Cleanroom and HVAC Qualification/Validation: Supports verification of room classification, airflow patterns, and filtration performance.

  • Personnel and Process Monitoring: Deployed during set-up, interventions, and routine work to assess operator- and process-associated shedding.

  • Excursion Investigations: Aids root-cause analysis and corrective action effectiveness checks after environmental monitoring alerts.

  • Facility Environmental Surveillance: Applied in pharma, biotech, medical device, hospital/compounding, and food/beverage areas to monitor ambient microbiological quality.

  • Recovery of Nonfastidious Microorganisms: General-purpose cultivation of airborne bacteria and fungi such as Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Bacillus spp.

  • Qualification of Cleaning and Disinfection Programs: Verifies the effectiveness of environmental cleaning regimens via fall-out monitoring.

  • Process Change Control: Used before/after changes to equipment, layout, or HVAC to assess impact on airborne bioburden.
