1 M Tris-HCl, pH 8.5 is a ready-to-use buffer stock used to establish and maintain alkaline pH in molecular biology and biochemistry workflows. The pH 8.5 setting is suitable for handling nucleic acids and for enzyme reactions that operate in mildly alkaline conditions.

Typical applications include dilution to prepare working-strength Tris buffers (e.g., 10–100 mM) for DNA or RNA resuspension, elution of plasmid DNA from silica matrices, preparation of Tris-buffered saline and related solutions, and formulation of electrophoresis buffers based on Tris. It is commonly used in plasmid preparation workflows from E. coli and in processing protein extracts where controlled pH is required. GMP manufactured.

Polymerase Chain Reaction (PCR): Maintains optimal pH in PCR and qPCR master mixes for polymerase activity.

Nucleic Acid Purification and Storage: Used to prepare TE buffer and DNA/RNA elution or resuspension buffers at pH ~8.0–8.5.
Molecular Cloning: Provides buffering in restriction digests, ligations, and competent DNA handling workflows.
Protein Purification: Buffers for ion-exchange and affinity chromatography, maintaining stable pH during binding and elution.
Enzyme Assays: General buffer for enzymatic reactions requiring pH 7.5–9.0 stability.
Tris-Buffered Saline (TBS/TBST): Preparation of wash and dilution buffers for Western blotting and ELISA.
Cell Lysis and Protein Extraction: Base for Tris-based lysis buffers (e.g., RIPA variants) to stabilize proteins during extraction.
General Buffer Preparation and pH Adjustment: 1M stock used to quickly set and maintain pH in laboratory buffers.
