Tris buffer at neutral pH is a zwitterionic primary amine buffer used for pH stabilization in biochemical and molecular biology workflows. It is commonly incorporated into electrophoresis systems (e.g., TAE/TBE), protein sample and transfer buffers, nucleic acid purification workflows, and chromatography fractions, and for general reagent dilution and cell lysis buffer preparation, including applications with E. coli lysates.

The buffering range centers around physiological pH, with a temperature-dependent pKa that can shift measured pH with changes in temperature. Tris solutions are compatible with common additives such as EDTA and salts. Store tightly capped to limit atmospheric CO₂ absorption and prepare with high-purity water to minimize ionic contaminants.

  • Restriction Digestion and Ligation: Serves as the buffering base for enzyme reaction mixtures to support endonucleases and ligases.

  • Western Blotting and Immunoassays: Used to prepare Tris-buffered saline (TBS) and transfer buffers for blotting, blocking, and washing steps.

  • Cell Lysis and Protein Extraction: Included in lysis buffers (e.g., Tris-based formulations) to maintain pH during protein solubilization.

  • Nucleic Acid Extraction and Plasmid Preparation: Used in buffers for plasmid isolation from Escherichia coli and other workflows.

  • Dialysis and Sample Desalting: Provides a stable pH reservoir for buffer exchange and desalting of proteins and nucleic acids.
